If an incorrect nucleotide is in a DNA molecule due to mutation, will protien synthesis be affected?

Of course yes. thats bcoz every amino acid in the protein chain is coded by a particular codon consisting of three nucleotides in the messenger RNA. If the nucleotide of the DNA is changed then the whole codon would change and hence the amino acid.
